What is Kisspeptin-10?

Kisspeptin-10 (KP-10), also referenced in the literature as metastin (45–54), is a ten-residue, C-terminally amidated peptide with the sequence Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe-NH2. The terminal Arg-Phe-amide motif places it structurally within the RFamide peptide family, a group of C-terminally amidated signalling peptides sharing that same two-residue amide cap. Full-length kisspeptin is synthesised as a 145-amino-acid precursor and proteolytically processed into a series of shorter, biologically active fragments — kisspeptin-54, kisspeptin-14, kisspeptin-13, and kisspeptin-10 — all of which share this identical C-terminal decapeptide.

The kisspeptin family signals through KISS1R, also referred to in older nomenclature as GPR54, a Gq/11-coupled receptor that activates phospholipase C and downstream inositol trisphosphate and diacylglycerol signalling on binding. Longer fragments such as kisspeptin-54 resist this degradation pathway and persist longer in solution. Research context

Published research on kisspeptin peptides is concentrated on the hypothalamic-pituitary-gonadal axis. Study designs examine how KISS1R agonism on GnRH neurons — concentrated in hypothalamic regions such as the arcuate nucleus and the anteroventral periventricular nucleus in rodent models — influences the frequency and amplitude of pulsatile GnRH release, and how that pattern is reflected in luteinising hormone secretion from the anterior pituitary in rodent, ovine, and primate model systems.

Reconstitution

Direct the diluent stream against the inner wall of the vial rather than onto the powder itself, and allow the material to dissolve without agitation. Storage and stability

After reconstitution: refrigerate at 2–8 °C and protect from light. Reconstituted peptide in solution is materially less stable than the dry powder. Avoid repeated freeze-thaw cycles, which are a well-documented cause of aggregation and loss of measurable potency — aliquot instead if repeated sampling is required.

Inspect solutions before use. Cloudiness, visible particulate, or discolouration indicate the material should not be relied upon for quantitative work.
