What is ARA-290?

ARA-290 is not a fragment of erythropoietin’s linear sequence so much as a reconstruction of one surface of it. Erythropoietin (EPO) is a 165-amino-acid glycoprotein whose activity depends on the three-dimensional presentation of several discontinuous regions, one of which — the “helix B” surface — is reproduced here as a short, independently folded 11-residue peptide. Reducing a 165-residue protein to an 11-residue peptide is the structural basis for the compound’s distinct receptor profile relative to native EPO.

That distinct profile is described in the literature through the “innate repair receptor” (IRR) hypothesis. Classical erythropoietic signalling proceeds through a homodimer of the erythropoietin receptor, (EPOR)2. Reconstitution

Direct the diluent stream against the inner wall of the vial rather than onto the powder itself, and allow the material to dissolve without agitation. Storage and stability

After reconstitution: refrigerate at 2–8 °C and protect from light. Reconstituted peptide in solution is materially less stable than the dry powder. Avoid repeated freeze-thaw cycles, which are a well-documented cause of aggregation and loss of measurable potency — aliquot instead if repeated sampling is required.
