Semaglutide is a synthetic peptide analogue of human glucagon-like peptide-1 (GLP-1), structurally modified to resist enzymatic degradation and to bind reversibly to serum albumin. AminoOptimization Group supplies it as a lyophilised research powder in 5 mg, 10 mg and 20 mg presentations, tested to 99% HPLC purity, with a Certificate of Analysis available for the lot supplied. This material is offered strictly for laboratory research use. It is not a drug, and it is not for human or veterinary use.

What is SEMAGLUTIDE?

Semaglutide is a 31-amino-acid peptide sharing approximately 94% sequence identity with native human GLP-1(7-37). Two substitutions distinguish the backbone from the endogenous sequence: alpha-aminoisobutyric acid (Aib) in place of alanine at position 8, and lysine in place of arginine at position 34. The Aib8 substitution is described in the literature as conferring resistance to cleavage by dipeptidyl peptidase-4, the enzyme principally responsible for the short circulating half-life of native GLP-1.

The lysine introduced at position 34 carries a side-chain modification: a linker built from two units of 8-amino-3,6-dioxaoctanoic acid and a glutamic acid spacer, terminating in a C18 fatty diacid. This diacid group is the structural basis of the molecule’s non-covalent, reversible association with serum albumin — an approach shared conceptually with other fatty-acid-modified GLP-1 analogues, though the specific chain length and linker chemistry here are distinct.

That extended diacid and dual-linker arrangement is what separates semaglutide, as a chemical entity, from earlier albumin-binding GLP-1 analogues such as liraglutide, which carries a single C16 fatty acid attached through a single glutamic acid spacer. The compounds are studied in the literature as structurally distinct molecules, not as interchangeable variants of one another.

Research context

Published research on GLP-1 receptor agonists as a class examines receptor binding kinetics, the intracellular signalling that follows receptor engagement, and how the fatty-diacid and linker chemistry of a given analogue affects its albumin-binding affinity and clearance rate. This work is carried out in receptor-expression cell systems and animal models, and it is directed at characterising the compound’s chemistry and receptor pharmacology.

None of this literature establishes safety or efficacy for use in humans. It is provided as research context only.

Reconstitution

Lyophilised peptide requires reconstitution with a suitable sterile diluent before use in solution. Bacteriostatic water is typically specified where a preparation will be accessed more than once; sterile water is more common for single-use work.

Add the diluent slowly against the inside wall of the vial rather than directly onto the powder, and allow the material to dissolve on its own. Do not shake — peptides are sensitive to shear stress, and shaking can disrupt the folded structure. A gentle swirl is usually sufficient, and full dissolution at room temperature typically takes only a few minutes.

The concentration of the resulting solution depends on how much diluent is added to a given vial mass. Our peptide reconstitution calculator computes the resulting concentration for any combination of vial size and diluent volume.

Storage and stability

Before reconstitution: keep the sealed vial frozen at -20 °C and protected from light. In lyophilised form the peptide is comparatively stable, and brief temperature excursions during shipping are normal and are not expected to compromise a properly freeze-dried product.

After reconstitution: refrigerate at 2–8 °C and keep protected from light. Peptide in solution is materially less stable than the dry powder. Repeated freeze-thaw cycling is a well-documented cause of aggregation and loss of measurable material — aliquot the solution instead if it will be sampled repeatedly.

Inspect any solution before use. Cloudiness, visible particulate, or discolouration indicate the material should not be relied upon for quantitative work.

Certificate of Analysis

Each lot is tested by high-performance liquid chromatography, and the purity figure given above reflects the analytical result for the specific lot supplied, not a generic specification. A Certificate of Analysis is available on request, and lot-specific documentation can be provided before purchase where a protocol requires it.

Purity by HPLC is a statement about the proportion of target peptide relative to detectable related substances in the material as manufactured and tested. It is expressly not a representation that the material is suitable for any use in humans or animals.

Handling and safety

Handle in accordance with good laboratory practice. Use appropriate personal protective equipment, work within a suitable containment environment, and dispose of the material and any associated consumables according to your institution’s requirements and applicable local regulation. Anyone with access to this material should understand that it is not for human or veterinary use.

Frequently asked questions

What distinguishes semaglutide from other fatty-acid-modified GLP-1 analogues?

The C18 fatty-diacid group and the dual-linker chemistry connecting it to the peptide backbone are specific to this compound. Other GLP-1 analogues described in the literature use different acyl-chain lengths and linker arrangements, which published work associates with differences in albumin-binding affinity and clearance. These are studied as distinct chemical entities, not variants of one another.

Is semaglutide approved in Canada?

Semaglutide is the active ingredient in certain approved prescription medicines used under medical supervision. That regulatory approval belongs to those specific, manufactured pharmaceutical products — it does not extend to this listing. This research material has not itself been evaluated or authorised by Health Canada, the FDA, or any comparable regulatory authority; it carries no Drug Identification Number (DIN) or Natural Product Number (NPN), has not been assessed for identity, purity, or safety as a drug substance, and is supplied strictly for laboratory research use. It is not sold, represented, or intended as a substitute for, or equivalent to, any approved pharmaceutical product.

How much diluent should be used?

That depends on the working concentration a given protocol calls for. The peptide reconstitution calculator computes the resulting concentration for any combination of vial size and diluent volume.

How long is reconstituted material usable?

Peptide in solution is less stable than lyophilised powder, and how long it remains usable depends on diluent, temperature, and handling. Refrigerate at 2–8 °C, protect from light, avoid freeze-thaw cycling, and validate stability under your own storage conditions if your work depends on it.

Do you provide a Certificate of Analysis?

Yes — lot-specific, available on request, including before purchase.
