SNAP-8, referenced in the literature as Acetyl Octapeptide-3, is a synthetic octapeptide built on the same SNAP-25-derived fragment used in earlier hexapeptide analogues of this class, extended by two additional residues. AminoOptimization Group supplies it as a lyophilised research powder in a 10 mg presentation, tested to 99% purity by HPLC, with a Certificate of Analysis available for the lot supplied. This material is offered strictly for laboratory research use. It is not a drug, and it is not for human or veterinary use.

What is SNAP-8?

SNAP-8 is an eight-residue synthetic peptide — reported in the literature with the sequence Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Ser-NH2 — modelled on the N-terminal domain of SNAP-25, one of the three proteins that, together with syntaxin and VAMP/synaptobrevin, assemble into the SNARE complex responsible for vesicle docking and fusion at presynaptic membranes. Peptides built on this fragment are studied as short competitive mimics capable of occupying binding sites that are otherwise used during SNARE complex assembly.

SNAP-8 shares its core sequence with Acetyl Hexapeptide-8, a related hexapeptide analogue of the same SNAP-25 fragment, extended by two additional residues at the C-terminus. Published characterisation of the octapeptide describes a broader affinity profile than the shorter hexapeptide, with interaction reported to extend to additional components of the SNARE complex rather than SNAP-25 alone. This structural difference is the basis for comparative work examining whether the extended chain changes the kinetics or extent of complex interference relative to the six-residue analogue.

As a synthetic peptide rather than a recombinant protein, SNAP-8 is produced by solid-phase synthesis and purified by preparative HPLC. The N-terminal acetylation and C-terminal amidation present in the reported sequence are typical of peptides in this structural class and are described in the literature as contributing to resistance against exopeptidase cleavage relative to an unmodified linear peptide of the same length.

Research context

Peptides derived from the SNAP-25 N-terminal fragment are studied primarily for what they reveal about SNARE complex assembly and regulated exocytosis. Published research examines how competitive occupancy of SNAP-25 binding sites by a short synthetic fragment affects complex formation, and how this is reflected in measurable neurotransmitter and catecholamine release in secretory cell preparations such as chromaffin and PC12 cell models.

SNAP-8 appears in study designs comparing octapeptide and hexapeptide analogues of this fragment, since the additional residues are reported to alter affinity for SNARE-complex components beyond SNAP-25 alone. It is also examined in in vitro dermal models, including reconstructed skin and fibroblast contraction assays, as a tool for probing the neuromuscular-junction-adjacent signalling pathways implicated in dermal contractile behaviour.

None of this literature establishes safety or efficacy for use in humans. It is provided as research context only.

Reconstitution

Lyophilised peptide must be reconstituted with a suitable sterile diluent before it can be used in solution. Bacteriostatic water, which contains a preservative permitting multiple withdrawals, is the diluent most often specified for multi-use laboratory preparations; sterile water without a preservative is more typical where a single-use preparation is required.

Direct the diluent stream against the inner wall of the vial rather than onto the powder itself, and allow the peptide to go into solution without agitation. Peptides are sensitive to shear stress, so do not shake the vial — swirl gently if needed, and expect full dissolution within a few minutes at room temperature.

The concentration of the resulting solution depends on how much diluent is added to a given mass of peptide. Our peptide reconstitution calculator computes the resulting concentration for any combination of vial size and diluent volume.

Storage and stability

Before reconstitution: store the sealed vial in a freezer at -20 °C, protected from light. Lyophilised peptide is comparatively stable in this state, and brief excursions to ambient temperature during shipping are normal and are not expected to compromise a properly lyophilised product.

After reconstitution: refrigerate at 2–8 °C and protect from light. Peptide in solution is materially less stable than the dry powder. Avoid repeated freeze-thaw cycles, which are a well-documented cause of aggregation and loss of measurable peptide content — aliquot instead if repeated sampling is required.

Inspect solutions before use. Cloudiness, visible particulate, or discolouration indicate the material should not be relied upon for quantitative work.

Certificate of Analysis

Every lot is tested by high-performance liquid chromatography, and the purity figure shown above refers to analytical results for the specific lot supplied, not to a generic specification. A Certificate of Analysis is available on request, and we will provide lot-specific documentation before purchase if your protocol requires it.

Purity by HPLC describes the proportion of the target peptide relative to detectable related substances. It is a statement about the material as manufactured and tested. It is expressly not a representation that the material is suitable for any use in humans or animals.

Handling and safety

Handle in accordance with good laboratory practice. Use appropriate personal protective equipment, work in a suitable containment environment, and dispose of material and consumables according to your institution’s requirements and applicable local regulation. Anyone with access to this material should understand that it is not for human or veterinary use.

Frequently asked questions

What is the difference between SNAP-8 and Argireline (Acetyl Hexapeptide-8)?

SNAP-8 extends the same SNAP-25-derived sequence used in Acetyl Hexapeptide-8 by two additional residues. This longer chain is reported to broaden the peptide’s affinity to additional SNARE-complex components beyond SNAP-25 alone, which is why the two analogues are frequently studied alongside each other in comparative work.

Is SNAP-8 approved in Canada?

No. This compound has not been evaluated or authorised by Health Canada, the FDA, or any comparable regulatory authority. It carries no DIN or NPN, its safety and efficacy for human use have not been assessed, and it is supplied for laboratory research only.

How much diluent should be used to reconstitute SNAP-8?

That depends on the working concentration your protocol requires. Use the reconstitution calculator to work out the resulting concentration for your vial size.

How long is reconstituted SNAP-8 stable in solution?

Stability in solution is shorter than in lyophilised form and depends on diluent, temperature, and handling. Refrigerate at 2–8 °C, protect from light, avoid freeze-thaw cycling, and validate stability for your own storage conditions if your work depends on it.

Do you provide a Certificate of Analysis for SNAP-8?

Yes — lot-specific, available on request, including before purchase.
