What is BPC-157?

BPC-157 is a chain of 15 amino acids — Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val — identified as a partial sequence within body protection compound, a larger peptide originally isolated from human gastric juice. Four of the fifteen residues are proline, three of them consecutive at positions three through five. Structural work attributes much of the sequence’s resistance to proteolytic cleavage to this proline density, since proline’s cyclic side chain limits access for common gastrointestinal and serum peptidases.

This resistance to degradation is also what separates BPC-157 from most peptide fragments derived from tissue extracts: published in vitro work on the parent BPC sequence reports measurable stability across an extended incubation period in simulated human gastric juice, a property attributed to its amino acid composition. Unlike GHRH- or GHRP-class analogues, BPC-157 does not act through a defined pituitary secretagogue receptor. Its molecular target has not been established with the same certainty, and published literature continues to examine candidate binding partners.

Research context

Published research on BPC-157 concentrates on gastrointestinal and musculoskeletal tissue models. In vitro work examines the compound’s effect on fibroblast migration rates in cell culture, and rodent studies have investigated markers of angiogenesis, including VEGFR2 receptor expression and downstream signalling, in models of induced tissue injury.

A separate body of literature investigates BPC-157’s interaction with the nitric oxide system, examining nitric oxide synthase expression and NO-pathway activity in isolated tissue and animal models. The compound also appears in rodent study designs involving models of induced colitis and gastric mucosal injury, where gastrointestinal tissue markers are used as endpoints to examine proposed mechanisms of action.

Reconstitution

Reconstitution applies only to the 5 mg and 10 mg lyophilised vial presentations. The 500 mcg capsule format is a solid oral dosage form and does not require reconstitution or any diluent.

For the lyophilised vials, bacteriostatic water is the diluent most commonly specified for multi-use laboratory preparations; sterile water is typical for single-use work. Direct the diluent stream against the inner wall of the vial rather than onto the powder itself, and allow the material to dissolve without agitation. Peptides are susceptible to shear stress — do not shake the vial. Swirl gently if needed, and expect full dissolution within a few minutes at room temperature.

Storage and stability

Lyophilised vials, before reconstitution: store at -20 °C, protected from light. Lyophilised peptide is comparatively stable in this state, and brief excursions to ambient temperature during shipping are normal and are not expected to compromise a properly lyophilised product.

Lyophilised vials, after reconstitution: refrigerate at 2–8 °C and protect from light. Reconstituted peptide in solution is materially less stable than the dry powder. Avoid repeated freeze-thaw cycles, which are a well-documented cause of aggregation and loss of measurable potency — aliquot instead if repeated sampling is required. Inspect solutions before use; cloudiness, visible particulate, or discolouration indicate the material should not be relied upon for quantitative work.

Capsules: store in the original container at room temperature, away from moisture, heat and direct light. Reconstitution and freeze-thaw stability considerations for peptide solutions do not apply to it.
