What is ADIPOTIDE?
ADIPOTIDE is built from two functional domains joined by a short peptide linker. The first is a cyclic, disulfide-constrained targeting sequence originally identified through phage display screening, reported in the literature to bind prohibitin, a protein expressed on the luminal surface of vascular endothelial cells supplying white adipose tissue. The second is D(KLAKLAK)2, a synthetic amphipathic peptide characterized in earlier work for its capacity to disrupt mitochondrial membranes once it reaches the cytosol of a cell.
Neither domain is described as active in isolation the way the fusion construct is. (KLAKLAK)2 alone lacks cell-type selectivity — it disrupts membranes wherever it gains cytosolic access, which is why it is not studied as a standalone agent. Conjugation to the prohibitin-binding domain is described as restricting internalization, and by extension the pro-apoptotic effect, to the cell population reached by the targeting sequence. This ligand-directed design places ADIPOTIDE alongside other targeted pro-apoptotic peptides used in vascular-ablation research, distinct from a conventional receptor agonist or antagonist because the intended mechanism is localized cell death rather than modulation of a signalling pathway.
ADIPOTIDE should not be confused with prohibitin-binding peptides developed for other targeting contexts, nor with unconjugated (KLAKLAK)2, which is a separate research reagent with no targeting domain and a correspondingly different risk and specificity profile in the literature.

Research context

Reconstitution

After reconstitution: refrigerate at 2–8 °C and protect from light. Reconstituted peptide in solution is materially less stable than the dry powder. Avoid repeated freeze-thaw cycles, which are a well-documented cause of aggregation and loss of measurable potency — aliquot instead if repeated sampling is required.
